TheAntosgroupresearchestissueregeneration.Tounderstandthebiologyofregeneration,Dr.Antosisinterestedinansweringthreefundamentalquestions:
1.Howarecellsatthesiteofinjuryinducedtoregeneratelosttissues?
2.Howarecellsinvolvedinregenerationcontrolledtoproducethecorrectpattern?
3.Whatstopsregenerationoncetheappropriatesizeisreached?
Thesequestionsarehighlyrelevanttotissuebioengineeringandtothestemcellbiologyofrepairorreconstructionofhumantissues.TheAntoslabusesthezebrafishtoanswerthesequestions,becausethezebrafishwillregeneratemanyofitsorgans,includingheartandappendages.Therefore,thelabcandescribeanddissectthecellandmolecularmechanismsinvolvedinthefish’sregenerationabilitiesandrelatethemtothelimitsonmammalianregeneration.
AppendageRegeneration:
Althoughthezebrafishfinshavedifferentarchitecturestothemammalianlimbs,theycontainalmostallthesametissuetypes(bone,mesenchyme,joints,skin,melanocytes,etc.).Thesetissuesjustarearrangedinadifferentpattern.Unlikemammalianlimbs,afterpartialloss,thezebrafishfinwillregeneratecompletely.Becausethezebrafishcanberesearchedwiththecurrentcell,geneticandmolecularresearchtools,thisregenerationmodelallowstheAntoslabtoassessthemechanismsinvolvedinhowtostart,patternandstopthestemandprogenitorcellsinvolvedinregeneratingappendages.
HeartRegeneration:
HealthyheartphysiologyrequirescoordinatedCa2+handlingacrossthesyncytialcardiacmuscle.Whilesingle-cellcardiomyocyteexperimentsincultureindicatehowindividualcardiomyocytesaltertheirCa2+subcellulardistributionandhandlingproperties,theyarelimitedinaddressingthesyncytialnatureofthecardiomyocytesintheheartinvivo.Thus,experimentaldissectionattheorgan,cellandmolecularlevelsareneededtounderstandhowknownandto-be-discoveredmechanismsregulatecardiacphysiologyduringheartregenerationinvivo.TheAntoslabisthereforeusingallthecurrentcellandmolecularmethods,includingreal-timeinvivozebrafishheartimaging,toaddressfundamentalquestionsaboutcardiacinjury,regenerationandfunction.
