Our research is focused on the use of spectroscopic techniques, including deep-UV resonance Raman, to study protein structure, folding and dynamics. Selective excitation of the polypeptide backbone amide by UV light increases the intensity of the structurally sensitive amide modes in the UVRR spectra of proteins. We are primarily interested in using deep-UVRR spectroscopy to study protein-protein, protein-lipid and protein-small molecule interactions. Current projects include studies of lipid mediated folding of disordered (unfolded) peptides and the effect of oligomeric proteins on membrane integrity.
Multi-excitation UV resonance Raman (ME-UVRR) spectra represent a unique type of data that may be exploited to aide in the a priori deconvolution of the highly overlapped UV Raman bands of proteins. Current research projects involve developing robust chemometric methods for analysis of these multidimensional UVRR spectra for modeling peptide secondary structure, folding and conformational transitions.
The Chemistry Immersion Program (CHIP) is an educational outreach program for high school teachers and students aimed at increasing interest in chemistry and chemical measurement and spectroscopic technologies while improving students and teachers technology skill set.
