1. 真核细胞DNA复制起始机制：确定Chafer等新复制起始蛋白的生化性质和生物功能，建立体外DNA复制体系、阐明复制叉里的生化反应，研究复制起始严格调控机制；
2. 细胞调控维持停顿DNA复制叉稳定的分子机制：阐明一些新发现的checkpoint靶蛋白的调控机制、确定这些调控在维持停顿DNA复制叉稳定、维持基因组稳定的作用；
3. 真核细胞DNA单双链断裂修复机制：阐明新发现的DNA断裂修复蛋白作用机制，阐明乳腺癌易感基因BRCA1、BRCA2在DNA同源重组中的作用机制，确定PARP1的作用机制；
4）基本染色质结构形成机制：研究Chromsfork调控机制，研究基本染色质结构形成机制；
5）癌症药物筛选。
