In my Ph.D. studies as well as in my Postdoctoral fellowship I have been involved in the development and use of analytical chemistry methods directed to biologically relevant problems. The analysis of biomolecules by state-of-the-art techniques has taken me to a broad spectrum of fields, from plant analysis to proteomics experiments. As a postdoctoral fellow in the Metallomics Center of the Americas, Department of Chemistry, University of Cincinnati my research was focused on the regulatory effects of cytokines on the zinc proteome of macrophage in response to Histoplasma capsulatum infection, by analyzing through chromatographically coupled to elemental and molecular mass spectrometric techniques. The search for clinically relevant biomarkers has also been an area of interest, and has been applied to serum of hypertensive patients, in the search of metalloproteins and non-metal containing proteins as putative biomarkers for ischemic or hemorrhagic stroke. The focus of my current research is the role of metals in biological systems, in particular, the relevance of zinc, copper, iron and manganese in host-pathogen interactions. These chemical elements are essential micro nutrients for both pathogenic organisms and the host, and because of that, they can play an important role in the virulence or defeat of the invasive microorganisms. We have studied this interactions and found evidence of nutritional immunity, the directed deprivation of elements (Zn) as a defense mechanism of immune cells (macrophages), against pathogens at in vitro and in vivo models. By using metallomics schemes, we perform chromatographic separations with elemental specific ICP-MS or molecular structural ESI-IT-MSMS detectors to identify the metal containing species involved in this phenomenon. Our research interest extends beyond the nutritional immunity to the functional role of high and low molecular mass metal-containing species in the virulence of extracellular bacteria. This includes the signaling and activation of important excreted molecules, like proteases and linkage adherence factor proteins that can help the pathogens to avoid or survive the immune system attacks.
