研究领域

DNA紧密缠绕组蛋白形成核小体。核小体是染色质的基本重复单元,是表观遗传的物质基础。然而,核小体的组装和紧密染色质的形成阻碍DNA遗传信息的读取。DNA甲基化、组蛋白修饰、核小体排布以及异染色质结构等表观遗传机制是解码遗传信息的关键,在基因转录、细胞命运决定以及人类疾病等方面起着重要作用。 我们实验室主要以生物物理方法,结合生物化学以及其他手段,研究染色质调控因子的结构、工作机理以及生物学功能。我们的工作阐明了染色质重塑发生的机理,提出了染色质重塑的“DNA波”模型;我们解析了多个SWI/SNF家族超大染色质重塑复合物的高分辨结构,揭示其组装和识别核小体的机理,为相关基因突变的致病机理提供了理论框架;我们阐明了染色质重塑蛋白的活性调控机理,为染色质重塑蛋白应答染色质特征,调控染色质结构提供了理论框架。
