Mechanisms of regulated secretion from endocrine cells. Our primary focus has been on basic cellular and molecular mechanisms underlying vesicle trafficking and recycling pathways and intracellular 2nd messenger signaling in both anterior pituitary cells and pancreatic beta-cells. We are also investigating the cellular mechanisms that control glucagon secretion from pancreatic alpha-cells during hypoglycemia. All projects use a combination of techniques including mammalian cell culture, molecular biology and biochemistry, and quantitative single-cell imaging approaches including immunofluorescence, tracking of GFP-tagged proteins and ratiometric imaging of intracellular ion indicators. We use a variety of imaging technologies including digital-deconvolution combined with wide-field fluorescence and laser scanning confocal microscopy for FRAP and FRET of fluorescent protein-based probes. Projects have been supported by research grants from the National Institutes of Health (NIH), the Juvenile Diabetes Research Foundation International (JDRF) and the Barbara Davis Center for Childhood Diabetes.
