Transcriptional regulation of pro and anti-coagulant factors The main focus of current research is the transcriptional regulation of pro and anti-coagulant factors, in particular protein Z (PZ). PZ serves as a cofactor to protein Z-dependent protease inhibitor (PZI) in the inhibition of activated factor X (Fxa), a major procoagulant factor. The inhibition of FXa by ZPI is enhanced 1000-fold in the presence of PZ and PZ deficiency may lead to a mild bleeding tendency. FXa is not only involved in haemostasis but also inflammation and the cellular immune response, so PZ's role in inactivating FXa may also have implications for these processes. Human protein Z, like other procoagulant and anticoagulant factors, is synthesised in the liver and secreted into the blood. The organisation of the PZ gene and structure of the protein are very similar to those of coagulation factors factor VII, factor IX, factor X and protein C. The concentration of PZ in plasma has been found to vary widely between individuals. Because of this finding and the importance of PZ in the inactivation of Fxa, current studies are aimed at defining the transcriptional regulatory mechanisms of the PZ gene. Chemokine action in the endometrium In collaboration with Dr Susan Laird of the BMRC and T C Li of the Sheffield Teaching Hospitals' Jessops Wing ongoing work is looking to identify the role of chemokines in the endometrium. Microarray analysis and quantitative polymerase chain reactions (PCR) have been used to assess expression levels of potential target genes of chemokine signal transduction pathways. Further work to analyse the changes in expression of these potential targets should yield important insights into the endometrium.
