研究领域

利用多维基因组学数据的整合分析,研究人类重大疾病(肿瘤、老年疾病和遗传疾病等)的个体化诊疗策略,并进一步利用细胞系和模式生物解析增强子等调控元件的作用机制。 1. 从进化生物学的角度研究肿瘤内的异质性和肿瘤细胞的进化过程,探索新的癌症治疗模式。 2. 利用泛基因组数据,通过多维基因组学数据整合分析,在全基因组水平筛选与癌症发生发展紧密相关的增强子。以此为基础,结合CRISPRi-cas9技术,在多种癌细胞系中筛选与某种表型(增殖、迁移和抗药性等)紧密相关的增强子,进一步解析这些增强子的调控机制。以此为基础,探索新的癌症诊断和治疗策略。 3. 在全基因组水平筛选和挖掘参与成骨细胞分化的增强子和转录调控因子;结合GWAS、Hi-C和ChIP-Seq等多维基因组学数据,解析“增强子-转录因子-靶基因”调控路径;在细胞和小鼠模型中,利用CRISPR-cas9技术,解析候选增强子对成骨细胞分化调控的分子机制,并探索通过促进成骨细胞分化治疗骨质疏松的策略。 4. 利用家族谱系的外显子组(和全基因组)测序数据,解析罕见遗传疾病的新的致病基因,探索个体化诊疗模式。
