We are interested in developing in situ quantitative techniques that interrogate the folding, assembly, and misfolding of lipoprotein complexes that are the best molecular signatures of atherosclerotic risk. These will then be applied to attain mechanistic understanding that can lead to new therapeutic and diagnostic approaches for cardiovascular diseases. Ongoing projects include Integrating in situ analytical techniques with multiplexed quantitative proteomics to relate molecular proteostasis pathways to the properties of secreted lipoprotein particles. Molecular characterization of how physiologically relevant oxidation products induce aggregation and enhance atherogenicity of LDL Development of quantitative unbiased proteomic assays to both measure and characterize protein homeostasis in living cells. These will be applied to establish molecular etiologies and identify therapeutic targets for cardiovascular, neurodegenerative, and cancer diseases.
