研究领域

(1)睡眠调控的遗传基础与分子机制 睡眠的调控机制是睡眠领域长期以来的研究难点,确定调控睡眠的关键基因是睡眠领域竞争的焦点。本研究方向充分利用多种技术的优势互补,以基因修饰小鼠为模式动物,以不同层次的分子生化技术为手段,以睡眠调控为研究目标来展开相关科学实验。力争发现参与睡眠调控的关键效应基因,阐明睡眠调控的分子机制。 (2)重要睡眠功能的分子机制 短期或长期睡眠剥夺均会在不同的生物学层次上对生物体产生显著的负面影响,是探索睡眠功能的主要方法。(a)根据短期睡眠剥夺得到的结果,科学家先后提出了众多的理论假说,目前证据较为充足的是“记忆巩固说”和“突触稳态说”,其背后的分子机制是进一步研究的重要方向。(b)长期睡眠剥夺促使实验动物出现一系列特殊的生理现象,如食物摄取增加、体重减少、能量消耗增加、体温降低等,但是其背后的分子、生化、细胞等层次的机制还一无所知。本研究方向采用小鼠作为实验动物模型系统的研究长期睡眠剥夺改变小鼠生理特征的生物学机制,以期找到可应用于临床预防、治疗的靶标。 (3)睡眠研究新技术、新方法的开发与应用 融合其它相关学科的新技术和新方法是突破睡眠研究难点, 发现睡眠新功能的重要方式。本研究方向:(a)充分利用学校现有生物学研究技术平台和相关大型仪器,如光遗传学和双光子实时成像等,优化软件与硬件,进行睡眠调控与功能的相关研究;(b)充分利用现有睡眠动物模型,同我校其他领域的课题组进行合作,研究睡眠失调同不同重大疾病发生发展的相关性,如抑郁症和癌症等;(c)建设动物睡眠记录平台的开放式接口,将其它领域的先进技术同睡眠研究相结合,拓展睡眠领域的研究前沿。
