回国近5年来围绕STRIPAK超分子复合物,开展癌症与炎症信号机制研究,获得了前沿性成果:(1)解析STRIPAK复合物装配的基本结构特征,定义其整体不对称性以及激酶招募方式(论文以通讯作者发表在J Biol Chem,2014a;Cell子刊Structure,2013b;受邀发表综述在Curr Chem Biol, 2012);(2)系统揭示STRIPAK复合物激酶组分MST的活性调控机制,并探讨其在细胞生长和迁移中功能作用(论文以通讯作者发表在J Biol Chem,2014b;J Struct Biol, 2014;Cell子刊Structure,2013a;受邀发表综述在Cell Mol Immunol, 2012);(3)发现STRIPAK激酶组分MST信号通路(Hippo通路)中原癌蛋白YAP的一个天然拮抗蛋白VGLL4,解析三维结构,揭示功能机制,并设计发展了YAP的靶向抑制剂,为针对YAP/VGLL4比率失调的癌症提供一种新的个性化诊疗策略及途径(论文以通讯作者发表在Cancer Cell,2014,获Nature China专评;以共同通讯作者发表在Cell Res,2014;受邀发表综述在Acta Biochimica et Biophysica Sinica, 2014;以第一发明人申请专利1项)。
