(1)研究万向和研究简介:
①动物條细胞克隆与胚胎工程方向
从事山羊胚胎细胞和体细胞克隆的研究,1999年,与中国科学院发育生物学研究所合作,应用细
胞移植技术获得了体细胞克隆山羊,其成果获2000国家科技部十大科技新闻,目前已经建立了一
套完整的山羊州细胞克隆技术体系,先后以皮肤成纤维细胞、山羊乳腺上皮细胞及转基因体细胞
为供核细胞获得克隆羊,至今出生了60多只体细胞克隆羊;
②动物乳腺生物反应器研究方向
转基因山羊和家兔乳腺生物反应器和生物制药研究,先后获得:人乙肝病毒表面抗原(HBsAg)
、人促红细胞生成素(HEP0)、重组人纤溶爾原激活剂(rhAP)、人白介素-2(hIL-2)乳腺特
异性表达转基因羊或兔。部分成果已经进入开发应用阶段,并建立了重组蛋白提取、纯化及制剂
技术;
③转基因和基因编辑动物生物新品种培育研究方向
利用胚胎显微注射和动物体细胞克隆技术进行转基因生物新品种培育的研究,成功制备了人乳铁
蛋白(hLF)、人溶菌酶(hLYZ)等转基因山羊,并进入了生物安全中间试验;近年来应用
CRISPR/Cas9和TALE瞰术进行哺乳动物个体基因编辑研究,成功地获得了MSTN基因编辑兔和山
羊,创制高产优质动物新品系;
④转基因和基因编辑动物模型研究方向
从事转基因动物模型的制备,获得了复制型HB儼基因小鼠。目前正在进行高血脂和心血管病基
因编辑兔和APOE、LDLRM因敲除兔模型研究,已经建立了兔、羊基因编辑技术平台。
