General Fmoc‐Based Solid‐Phase Synthesis of Complex Depsipeptides Circumventing Problematic Fmoc Removal

Fmoc‐based solid‐phase syntheses of depsipeptides are often hampered by side‐reactions arising from the Fmoc removal step. This study explores the best conditions to circumvent such drawbacks and the findings are used to prepare a highly complex linear depsipeptide exclusively on solid phase. This approach opens the door to a general fully Fmoc‐based strategy for the synthesis of complex depsipeptides.
