Three homologous metalloenzymes—tyrosinase, TYRP1 and TYRP2—catalyze the biosynthesis of melanin in humans, a pigment that is responsible for the pigmentation of skin, eye, and hair. The recently determined crystal structure of TYRP1 provides a structural rationalization of oculocutaneous albinism type 3 related mutations, and gives insight into binding modes of substrates and inhibitors that can guide structure-based design of compounds for pigmentation disorder treatments. This review discusses the current knowledge on the activity and function of the three melanogenic enzymes.
