A versatile theranostic nanosystem (FLAGC) is reported for rapid, non-invasive, accurate diagnosis and efficient clearance of metastatic lymph nodes. FLAGC is easily drained to sentinel lymph nodes (SLNs) for precise localization. FLAGC is then specifically lit up by the low pH and over-expressed myeloperoxidase (MPO) in the tumor microenvironment (TME), resulting in an accurate diagnosis of metastatic SLNs by chemical resonance energy transfer (CRET) luminescence in the second near-infrared window. Further, the tumor was efficiently eradicated through Ce6-mediated photodynamic therapy (PDT). Our findings offer a new paradigm for identifying and eliminating clinically positive lymph nodes.
