Archaerhodopsin 3 is an Ideal Template for the Engineering of Highly Fluorescent Optogenetic Reporters

We are grateful to the reviewers for their globally positive evaluation and the very constructive criticism and suggestions they made. In particular, we acknowledge their request for a detailed discussion of the lessons the community can learn in view of devising new and better performing fluorescent mutants of AR-3. We have now addressed this question in a new section entitled "Engineering Aspects". The second major request from reviewer 1 concerned the conclusion section, which should focus on highlighting the outcome and possible implications of our work for future research in this area. In response to this, we have fully re-organised the conclusion section and moved the paragraphs discussing and summarising the main results to the relevant sections 2 and 3. Other items and questions raised by the reviewers were addressed in detail and led to minor changes (see detailed point-by-point response). In conclusion, we hope that the new version convinces the reviewers, and that the manuscript will be accepted for publication in Chemical Sciences.
