A DNA nanotechnology-based strategy for spatially assembling heterotypic antigens (hetero-POINT) has been developed to rationalize the design of mosaic nanoparticle vaccines. Immunological investigations reveal that the pattern of heterotypic antigen distribution yields distinct humoral and mucosal immune effects, highlighting the significance of the spatial placement of the antigens in structure–activity relationships.
