A convenient methodology for the preparation of 6-functionalized azabicyclo[4.1.0]heptane-derived building blocks is developed. The common synthetic precursor – tert-butyl 6-(hydroxymethyl)-3-azabicyclo[4.1.0]heptane-3-carboxylate was obtained in 4 steps starting from readily available 4-hydroxymethyl pyridine. Other 6-functionalized and 3,6-difunctionalized azabicyclo[4.1.0]heptane-derived building blocks (including novel bicyclic γ-amino acid) were prepared in short synthetic pathways through 1–3 steps.
