Starch from Pulses Has Prebiotic Effects on Healthy Human Donors in an In Vitro Study

Pulse starch (PS) contributes to the resistant properties associated with the consumption of pulses. Since starch contributes to the resistant starch (RS) properties of pulse seeds, we hypothesized that PS alone has prebiotic effects. Upper GI digestion was performed on PSs in sodium phosphate buffer (pH 6.9) for 3–5 min, cooled (RT, 30 min), dialyzed (6–8 kDa, 36 h, and 6 h change of water interval), and lyophilized (72 h). Upper GI digestion resulted in 11–13% RS in all the investigated PSs, promoting acetate and butyrate production. Further, 16S sequencing results revealed that all the RSs significantly increased Bacteroidetes while significantly decreased Firmicutes. At the Phyla level, RS from PS significantly promoted Bacteroide, Faecalibacterium, Ruminococcus, and Blautia. Our findings showed that PS has RS from PSs modulating gut microbiota composition, facilitating PS-RS prebiotic potential.
