Megasynthases are responsible for producing primary metabolites and complex natural products. The final products are built in a stepwise process in which small acyl carrier proteins (ACPs) play a central role as they shuttle the growing intermediates between the different catalytic domains. This Review provides an overview of the current understanding and proposes a mechanism for the programming of the ACP movement in megasynthases.
