Self-immolative amphiphilic poly(ferrocenes) are fabricated with self-immolative backbone, aminoferrocene (AFc) side chains, and end-capping poly(ethylene glycol) monomethyl ether. Upon acidic milieu-triggered head-to-tail degradation, rational GSH depletion and reactive oxygen species burst is observed to amplify the oxidative stress for efficient tumor inhibition. This work provides a promising drug-free polymer strategy for precision medicine.
