Efficient Production of 3-Amino-2-Hydroxy Acetophenone by Multi-Enzyme Biosynthesis

We developed a biosynthetic route from m-nitroacetophenone (1a) to 3-amino-2-hydroxy acetophenone (1c) via 3-hydroxy amino acetophenone (1b) using a direct reaction method with three enzymes and supersaturated substrates. Through enzyme molar ratios, expression, and structure optimization, we obtained the optimal mutant with a biosynthetic yield of 580 mg/L.
