The 3D structure of cyclobutane opens opportunities for unique applications in medicinal chemistry. Improved synthetic methods and commercial availability now allow for more facile incorporation of this structural motif in small-molecule drug candidates. Although its use has been increasing, it is still very much underused relative to other motifs in drug development. This review presents recent applications of the cyclobutane ring in small-molecule drug development and the utility it can offer to medicinal chemists.
