Asparaginyl endopeptidase in developing and germinating legume seeds: immunological detection and quantitation

Using an antibody raised against jackbean pro-asparaginyl endopeptidase and a substrate for jackbean asparaginyl endopeptidase, immunoblot analysis and quantitation of the activity were performed for extracts from cotyledons of both developing and germinating seeds of five legume species, jackbean, soybean, Vigna mungo, peanut and kidney bean. The immunoblot analysis indicated the occurrence of multiple polypeptides immunologically related to jackbean asparaginyl endopeptidase in each legume examined. Most of the polypeptides detected in developing and dry seeds had different molecular masses from those in germinating seeds. Asparaginyl endopeptidase activity as measured by using dinitrophenyl(DNP)-Pro-Glu-Ala-Asn-NH2 as a substrate was detected in extracts from developing and germinating legume seeds tested. A possible role of this asparaginyl endopeptidase activity during seed germination is discussed.
