To overcome the delivery and cell uptake hurdle to siRNA therapeutics, modifications have been made to improve in‐vivo delivery through numerous methods. Functional group modifications on ribose rings, alterations in phosphate backbone linkage, and conjugation of GalNAc, lipid and disulfide ligands to the siRNA duplex are all potential contributors to improving current gene therapies.
