Mr. Sun gave the following suggestions for such problems: 1. Inhibitor preservation, preparation of mother liquor (stock solution): must be centrifuged, determine the solvent and maximum solubility, if insoluble, can be water bath (40-50 degrees, within 30 minutes) or ultrasonic to clarify, aliquot storage, avoid repeated freeze-thaw. 2. Solvents and solubility. 3. Dilution of inhibitor mother liquor: dilution solvent is recommended to use culture solution, mother liquor concentration according to the experiment to customize, generally more than a thousand times the concentration of working solution, gradient dilution, if the single dilution ratio is too large, can use secondary dilution (100 * 100). Later, Mr. Sun said that Selleck used elemental analysis to determine the actual content of inhibitors, which can ensure the authenticity of drug concentrations.
