      结直肠癌(CRC)是一种常见的且致命的恶性肿瘤。一些研究发现,CRC中存在的细菌Fusobacterium nucleatum (Fn,具核梭杆菌)具有促进肿瘤发展和增强肿瘤化疗耐药型的效果,使CRC的实际治疗效果受到影响。此外,由于细菌生物膜形成等问题,传统抗生素给药难以有效清除肿瘤部位的Fn,导致肿瘤化疗效果降低。因此,开发清除Fn和提高抗肿瘤疗效的新方法对改善CRC治疗具有重要意义。本研究开发了一种整合了油酸修饰的超顺磁性氧化铁纳米粒子(O-SPIONs)和两亲性聚合物(PPL)的纳米药物OPPL,用于递送铂类前体药物和月桂酸(LA,抗微生物),增强CRC的治疗效果。OPPL通过类过氧化物酶产生ROS,从而协同增强OPPL对Fn和LA的抗菌及生物膜清除活性。此外,OPPL纳米药物还可以促进细胞内ROS产生,增强细胞内脂质过氧化和谷胱甘肽耗竭,从而导致细胞铁死亡。联合化疗和诱导铁死亡策略使OPPL纳米药物对CRC细胞表现出较高的细胞毒性。体内研究表明,OPPL纳米药物可以增强肿瘤药物聚集,实现磁共振成像,抑制肿瘤生长及瘤内Fn的生长。本研究结果表明,OPPL治疗Fn感染CRC的效果优良,是一种有前景的CRC治疗候选药物。
