Addressing the four challenges: New concepts for C−H activation and catalyst design 1. Weakly coordinating substrates which recruit and position the Pd catalyst for C–H activation whilst avoiding the scope limitations associated with strongly binding directing groups. 2. Distance and geometry can be rationally tuned and employed as the key element to differentiate multiple remote C–H bonds with highly similar electronic and steric properties, allowing the control of site selectivity through templated macrocyclic transition states. 3. Bifunctional ligands (e.g. mono N-protected amino acids) which actively participate in the C–H cleavage transition state, thus enabling reactivity through ligand acceleration and imbuing C–H activation selectivity through providing a defined chiral environment at the enantiodetermining step. These ligands also have allowed the use of practical oxidants such a peroxides and molecular oxygen for catalytic turnover. The discovery of ligand-supported Pd catalysis marks a significant departure from early studies using simple Pd salts for C–H metalation. Since 2008, several generations of bifunctional ligands have been developed in our laboratory: MPAA, MPAThio, MPAAM, MPAQ, MPAO, Pyridone, Pyridone-Pyridine.
