研究领域

天然药物碳骨架的生物合成与编辑对惰性碳的选择性活化是化学领域的难点和热点，亦是药物研发和生产领域的瓶颈。天然产物成药高度依赖于结构优化，然而由于骨架碳原子反应惰性及整体结构的复杂性，对天然产物碳骨架的结构修饰极具挑战，因此复杂天然产物碳骨架的成药性研究一直被认为是天然药物研发的“禁区”。对于结构相对简单的合成/半合成药物的生产，由于缺乏经济的、高立体选择性的骨架合成及直接的碳位点延伸方法，常需繁冗的反应步骤和高昂的生产成本实现药物的合成/半合成。因此，普适、高效的碳骨架修饰/合成方法，对天然药物研发和药物生产具有重要意义。聚酮、甾体和氮杂环涵盖了近7万种天然产物和一千多种天然/半合成/合成药物，是重要的化合物类群。本实验室长期致力于这三大类天然产物骨架的生物合成与编辑方法的开发与应用。通过解析天然产物骨架生物合成的普适性规律，综合酶工程，生物催化，结构生物学，合成生物学等多学科技术手段，对生物合成途径进行升级或再设计，发展出针对上述各类骨架的普适、高效的生物合成与编辑策略，用于拓展结构多样性和提高药物的生产效率，实现对三大类碳骨架资源及药物的深度开发和高效生产。
