研究领域

长期致力于生物生存策略及环境适应机制研究、探索有毒动物(如蜈蚣、牛虻等)捕食和防御策略,揭示生物环境适应的物质基础与分子机制、并基于天然活性物质进行新药研发 1)研究高原两栖动物环境适应机制,从臭蛙皮肤发现第三套抗氧化系统,对皮肤保护、衰老研究与药物研究应用具重要意义(MolCellProteomics2009,8:571-583),该工作得到德国哥廷根大学BurkhardPoeggeler教授高度赞赏“这是该领域的先驱工作,近期内没有任何其他工作在同领域有类似影响”; 2)研究两栖动物微生物防御机制,从铃蟾等识别了超过400种抗菌肽,占目前世界两栖动物抗菌肽总数40%以上,揭示两栖动物环境适应机制,也为抗感染药物研发提供源头资源(ChemRev.2015;115:1760-846),并利用抗菌肽作为工具研究动脉粥样硬化疾病机理(Immunity2015;43:1137-47); 3)研究树蛙对捕食者的防御策略,从华西树蛙发现神经毒素,这是首次发现的两栖动物基因编码的神经毒(JBiolChem.2009,284:22079-86),为镇痛新药研发提供了候选药物、并被美国生物化学与分子生物学联合会作为亮点工作重点报道及被ScienceDaily等二十多家国际媒体报道; 4)研究吸血节肢动物吸血策略,揭示牛虻从其宿主成功吸血分子机制,从牛虻唾液腺发现>100种抗血栓和抗感染多肽,为药物研发提供源头资源(MolCellProteomics.2008;7:582-90.MolCellProteomics.2009;8:2071-9);探索埃及伊蚊与宿主互作机制及吸血策略,发现其唾液腺成分通过抑制宿主免疫而协助寨卡病毒传播(NatureImmunology,19(4):342-353),并为自身免疫药物研发提供及源头资源; 5)研究蜈蚣捕食策略,首次发现钠通道NaV1.7专一抑制剂(PNAS2013;110:17534-9),被Nature、NatureMedicine进行专门评论,并被推为Faculty1000,认为是目前全世界发现的最好的治疗慢性疼痛候选药物;蜈蚣毒素成分SsTx阻断离子通道KCNQ从而实现对巨大猎物高效捕食(PNAS2018;115:1646-1651),该工作被Science、华盛顿邮报、德国新闻社等专门评论;蜈蚣通过其毒素成分激活辣椒素受体诱导捕食者产生剧烈疼痛,从而逃避捕食(NatCommun.2015;6:8297),为镇痛药物研发提供新思路和新的工具药物; 通过这些工作,揭示有毒动物环境适应策略;以通讯作者身份在Immunity(IF25)、NatureImmunology(IF22),PNAS(2篇),NatureCommunications,ChemicalReviews(IF50)等著名国际刊物上发表SCI研究论文170篇(具体论文情况可通过如下链接查询:https://www.ncbi.nlm.nih.gov/pubmed/?term=lai+ren),识别了自主知识产权活性多肽1300多种,提供候选药物资源,获发明专利授权60项,奠定了我们在该领域的国际领先地位,受邀在国际顶级刊物ChemicalReviews(IF50)发表长篇综述论文(ChemRev.2015;115:1760-846)、利用毒素多肽研发药物或工具药物(如一工具药物已在全球销售RHT-3832-PI,https://www.pepnet.com/Products/Detail/3766/Rh-Tx);利用有毒动物资源推动山区农村脱贫致富,十一五、十二五期连续两次被云南省评为“扶贫工作”先进集体。
