硒蛋白及稀土化合物与疾病发生、发展的关系及其抗老年痴呆症和抗癌的作用机制的研究(深圳大学) 多功能纳、微米粒子载体的合成及其生物医学应用研究(长春应用化学研究所) 研究领域 1. 硒蛋白及稀土化合物与疾病发生、发展的关系及其抗老年痴呆症和抗癌的作用机制的研究 研究硒蛋白的合成结构及与重大疾病的关系(如用生物信息学研究);不同物种中硒蛋白的预测;研究硒蛋白的克隆、表达及结构与功能;探索硒蛋白及稀土化合物与老年痴呆症、癌症等重大疾病的关系。硒在生物体内主要以硒蛋白形式参与调节各种生理环节、发挥不同生物功能,直接或间接地与多种重大疾病(癌症、神经退行性病变和病毒等)相关联。然而因硒蛋白基因结构的复杂性,故有关对重大疾病的机理研究尚很不深入,极大地限制了其应用。因此,对硒蛋白生物信息学预测,用基因工程表达及化学修饰等方法研究硒蛋白的结构与功能的关系,将为探索不同硒化合物作为预防药物提供基础数据,稀土化合物作为癌症药物尚有很大争议。故用蛋白质组学及代谢组学及细胞生物无机化学方法。研究稀土化合物与癌细胞的作用。将有助于澄清有关的争议。为稀土药用前景提供重要信息。 2.多功能纳、微米粒子载体的合成及其生物医学应用研究  针对癌症的发病机理、早期诊断和靶向治疗,设计合成出集荧光、磁性和特异识别于一体的新型多功能载体用于功能生物分子的富集分离检测、癌细胞的生物探针诊断和靶向控释药物治疗。该多功能载体同时具有选择富集、磁控操纵、荧光成像、磁共振成像和靶向药物控制释放等多种功能。基于该多功能载体,对蛋白质、酶、DNA等功能生物分子可进行高效选择富集、快速磁分离和高灵敏度检测;对外周血液中的癌细胞可实现特异识别和可视化捕获用于富集鉴别;对恶性肿瘤实体可进行荧光成像和磁共振成像检测;还可以通过药物靶向定位和控制释放对恶性肿瘤实现有效地治疗。在癌细胞的早期诊断和癌症的靶向药物治疗等领域具有极大的应用前景。
